Abstract: The present invention provides a portable microfluidic electrochemical biosensor system and method for quantitatively determining gamma-aminobutyric acid (GABA) in saliva. The system comprises a disposable cartridge having a sample inlet, passive capillary-driven microfluidic network, selective filtration membrane, immobilized dual-enzyme recognition layer and three-electrode sensing assembly comprising a nanostructured platinum-black/gold-nanoparticle working electrode. The enzyme layer converts GABA into hydrogen peroxide, which is electrochemically oxidized to generate a current proportional to the GABA concentration. A reusable portable reader comprising a potentiostat, signal-conditioning circuitry, analog-to-digital converter, processor and memory processes the current using digital filtering and calibration to determine the GABA concentration. The determined concentration is displayed and may be wirelessly transmitted to a mobile device for point-of-care assessment. The system provides rapid, non-invasive, sensitive and selective quantification of salivary GABA while improving analytical stability, reproducibility and portability.
