Abstract: The present invention discloses a novel polyherbal ethosomal nanoformulation comprising green synthesized silver nanoparticles incorporated with standardized Ashwagandha (Withania somnifera) extract and complementary herbal bioactives for enhanced burn wound management. The formulation utilizes phospholipid-based ethosomal vesicles containing ethanol to improve dermal penetration and sustained release of active phytoconstituents. Biogenic silver nanoparticles synthesized using plant extracts provide potent broad-spectrum antimicrobial activity against burn-associated pathogens including Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, and fungal contaminants while minimizing cytotoxicity. Ashwagandha extract supplies withanolides that exhibit anti-inflammatory, antioxidant, immunomodulatory, collagen-stimulating, and tissue regenerative activities. Additional herbal extracts such as Aloe vera, Curcuma longa, Centella asiatica, and Azadirachta indica synergistically promote wound contraction, angiogenesis, extracellular matrix remodeling, and epithelial regeneration. The ethosomal carrier enhances skin permeation, improves stability of phytochemicals, increases retention within damaged tissues, and provides controlled release of therapeutic agents. The formulation may be prepared as a gel, cream, spray, hydrogel, dressing, or impregnated wound patch suitable for topical administration. Experimental evaluation demonstrates significant reduction in microbial load, inflammatory cytokines, oxidative stress, wound size, and healing duration while improving collagen deposition, hydroxyproline content, and tensile strength compared with conventional burn therapies. The invention offers a safe, biocompatible, and cost-effective alternative for comprehensive burn wound management.
