Abstract: The present invention relates to a pharmaceutical composition comprising Berberine and Olanzapine for the treatment of breast cancer. More particularly, the present invention provides a synergistic combination of the natural bioactive compound Berberine and the repurposed pharmaceutical agent Olanzapine, which exhibits enhanced cytotoxic, anti-proliferative, and pro-apoptotic effects against breast cancer cells. The synergistic interaction between the components is characterized by a combination index (CI) of less than 1, indicating enhanced therapeutic performance compared with individual treatments. The present invention is supported by computational studies, including molecular docking and ADMET analysis, as well as experimental validation through in vitro assays such as the MTT assay, wound- healing assay, colony-formation assay, and apoptosis analysis. Accordingly, the present invention provides a cost-effective, clinically translatable, and efficient therapeutic strategy for improved management of breast cancer.
