Abstract: ABSTRACT TITLE: A PHARMACEUTICAL COMPOSITION COMPRISING ORYZANOL-FORTIFIED ETHOSOMAL GEL FOR MANAGEMENT OF RHEUMATOID ARTHRITIS AND PREPARATION METHOD The present invention relates to a pharmaceutical composition comprising an oryzanol-fortified ethosomal gel for the transdermal management of rheumatoid arthritis, and a method of preparation thereof. The composition comprises oryzanol encapsulated within nanosized phospholipid ethosomal vesicles containing phospholipid, cholesterol, ethanol and purified water, dispersed into a Carbopol-940 hydrogel base together with glycerol, propylene glycol and triethanolamine at a skin-compatible pH of 5.0 to 6.5. The ethosomes exhibited a mean vesicle size of about 205 nanometres, PDI of 0.116 and zeta potential of 39.27 mV. The gel demonstrated 84.63 per cent cumulative drug release over 24 hours, skin permeation of 49.9 micrometres, significant reduction of paw oedema, restoration of haematological parameters, and marked suppression of TNF-alpha and IL-6 in the CFA-induced arthritic rat model, offering a stable, scalable and effective natural transdermal therapy.
