Abstract: The present disclosure proposes a pharmaceutical composition (100) and a method for preparing a mucoadhesive gastroretentive tablet (300) of acotiamide hydrochloride (102). The mucoadhesive gastroretentive tablet (300) is configured to exhibit prolonged gastric residence, controlled drug release, and enhanced retention within the stomach through mucoadhesive and buoyancy mechanisms. The pharmaceutical composition (100) comprises the acotiamide hydrochloride (102), ice apple (Borassus flabellifer) mucilage (104), hydroxypropyl methylcellulose (HPMC K4M) (106), sodium bicarbonate (108), polyvinylpyrrolidone K30 (PVP-K30) (110), microcrystalline cellulose (MCC) (112), and magnesium stearate (114). The pharmaceutical composition (100) utilizes a natural, biocompatible, and biodegradable polymer derived from the ice apple mucilage (104) as a mucoadhesive agent, thereby reducing reliance on synthetic excipients. The pharmaceutical composition (100) is suitable for long-term administration owing to the use of pharmaceutically acceptable and naturally derived excipients. The pharmaceutical composition (100) exhibits consistent physicochemical properties, including acceptable hardness, friability, swelling behaviour, and uniform drug content.
