Abstract: The present invention discloses a novel withanolide-loaded polymeric nanoparticle formulation exhibiting enhanced neuroprotective activity validated through molecular docking studies. The formulation comprises Withanolide A isolated from Withania somnifera roots encapsulated within biodegradable poly(lactic-co-glycolic acid) nanoparticles prepared by nanoprecipitation or double emulsion solvent evaporation. The optimized nanoparticles exhibit a mean particle size of 156.4 nanometers, polydispersity index of 0.187, zeta potential of negative 22.6 millivolts, and encapsulation efficiency of 82.3 percent. The formulation demonstrates sustained drug release over 72 hours following Higuchi kinetics. Molecular docking reveals strong binding of Withanolide A with acetylcholinesterase (binding energy negative 9.8 kcal/mol) and NMDA receptor (negative 8.6 kcal/mol). The nanoparticle formulation exhibits 3.7-fold enhanced blood-brain barrier permeation and superior neuroprotection in hydrogen peroxide-induced neurotoxicity model compared to free drug, offering a promising therapeutic strategy for neurodegenerative diseases.
