Abstract: The present invention relates to a reverse phase high performance liquid chromatographic (RP-HPLC) method for quantitative estimation of Piracetam in bulk drug substances and pharmaceutical dosage forms. The method employs a C18 column having dimensions of 150 mm × 4.6 mm and particle size of 5 µm. Chromatographic separation is achieved using a mobile phase comprising potassium dihydrogen phosphate buffer adjusted to pH 3.0 and acetonitrile in a ratio of 80:20 v/v at a flow rate of 1.2 mL/min. Detection is carried out at 210 nm with an injection volume of 20 µL. Piracetam is eluted at a retention time of approximately 3.20 minutes. The method demonstrates excellent specificity, linearity over the concentration range of 20–140 µg/mL, accuracy, precision, robustness, sensitivity and solution stability. The method provides a limit of detection of about 0.35 µg/mL and a limit of quantification of about 1.05 µg/mL. The method is suitable for routine quality control, assay determination and quantitative analysis of Piracetam in bulk drug substances and pharmaceutical formulations.
