Abstract: ABSTRACT A Novel Process for the Preparation of 3-((1H-Benzo[d]imidazol-1-yl)methyl)-5-amino-1,3,4-thiadiazole-2(3H)-thione The present invention relates to a novel and efficient process for the preparation of 3-((1H-benzo[d]imidazol-1-yl)methyl)-5-amino-1,3,4-thiadiazole-2(3H)-thione. The process comprises hydroxymethylation of benzimidazole using formaldehyde to obtain 1-(hydroxymethyl)benzimidazole, followed by chlorination to produce 1- (chloromethyl)benzimidazole. In a parallel sequence, 5-amino-1,3,4-thiadiazole-2(3H)-thione is protected using di-tert-butyl dicarbonate to form a Boc-protected intermediate. The protected thiadiazole is subjected to N-alkylation with 1-(chloromethyl)benzimidazole in the presence of a base to yield a coupled intermediate, which is subsequently deprotected under acidic conditions to afford the target compound. The process provides improved yields, operational simplicity, and suitability for industrial scale-up.
