Abstract: The present invention relates to a novel plant-derived prebiotic culture medium for enhancing growth, viability and functional metabolite production of probiotic microorganisms. The culture medium comprises plantain pseudostem powder and millet powder as prebiotic substrates, optionally supplemented with a nitrogen source and mineral salts. The plantain pseudostem is processed by washing, slicing, controlled drying and milling, while selected millet material is dehulled and finely ground. The substrates are combined in predetermined ratios and formulated at a suitable concentration and initial pH before sterilization. A selected probiotic microorganism is inoculated into the medium and cultivated under controlled fermentation conditions. The plant-derived substrates provide fermentable carbohydrates, dietary fibre and resistant carbohydrate fractions that support microbial proliferation and metabolic activity. The medium facilitates production of functional metabolites including short-chain fatty acids while maintaining probiotic viability. The formulation is suitable for laboratory and industrial cultivation and may be utilized for preparation of probiotic supplements, functional foods, nutraceuticals, synbiotic formulations and related microbial products.
