Abstract: ABSTRACT The present invention relates to a novel polymeric nanoformulation comprising standardized fenugreek seed (Trigonella foenum-graecum L.) extract and mulberry leaf (Morus alba L.) extract for improving insulin resistance and glucose homeostasis in Type 2 diabetes mellitus. The botanical extracts are incorporated into biodegradable polymeric nanoparticles, preferably chitosan nanoparticles cross-linked with sodium tripolyphosphate. Fenugreek extract may be standardized for 4- hydroxyisoleucine, trigonelline, diosgenin and associated phytoconstituents, while mulberry leaf extract may be standardized for 1-deoxynojirimycin, flavonoids and phenolic compounds. The nanoformulation is designed to enhance phytochemical stability, provide controlled release and improve delivery of the incorporated bioactive constituents. The combined formulation provides complementary metabolic actions including improvement of insulin signaling and peripheral glucose uptake, enhancement of GLUT4-mediated glucose utilization, inhibition of intestinal carbohydrate-digesting enzymes, reduction of postprandial glucose elevation, and modulation of oxidative and metabolic stress. The invention further provides a process for preparation of said polymeric nanoparticles and pharmaceutical compositions comprising the nanoparticles in oral dosage forms. The formulation provides a standardized multi-target botanical nanocarrier platform for prevention or management of insulin resistance and associated metabolic abnormalities of Type 2 diabetes mellitus.
