Abstract: The present invention relates to a bilosomal vesicular carrier-based pharmaceutical composition comprising Dasatinib or a pharmaceutically acceptable salt thereof for enhanced transdermal delivery in the treatment of skin cancer. Dasatinib exhibits significant anticancer activity; however, its clinical application is limited by poor aqueous solubility, extensive first-pass metabolism, variable oral bioavailability, and systemic adverse effects. The invention provides nanosized Dasatinib-loaded bilosomal vesicles comprising one or more phospholipids, one or more bile salts, one or more non-ionic surfactants, cholesterol, and pharmaceutically acceptable excipients to improve skin permeation, localized drug deposition, and sustained drug release. The bilosomal formulation is prepared by a thin-film hydration technique followed by vesicle size reduction through homogenization and is incorporated into a pharmaceutically acceptable topical gel for transdermal administration. The pharmaceutical composition exhibits improved physicochemical stability, enhanced retention within skin tissues, prolonged localized therapeutic action, and reduced systemic exposure compared with conventional Dasatinib formulations. The invention is useful for the localized treatment of melanoma, basal cell carcinoma, squamous cell carcinoma, and other skin malignancies, providing an effective, non-invasive, and patient-friendly transdermal drug delivery system.
