Abstract: The present invention relates to novel benzothiazole–thiazolidine-2,4-dione–1,2,3-triazole hybrid compounds, pharmaceutical compositions thereof, and a method for their preparation for therapeutic applications. The disclosed compounds integrate benzothiazole, thiazolidine-2,4-dione, and 1,2,3-triazole pharmacophores into a single molecular framework to enhance biological activity against vascular endothelial growth factor receptor-2 (VEGFR-2). The compounds are synthesized through sequential Knoevenagel condensation, azidation, and copper(I)-catalyzed azide–alkyne cycloaddition reactions, followed by purification and structural characterization using spectroscopic and analytical techniques. The synthesized compounds are evaluated through molecular docking, VEGFR-2 enzyme inhibition, and in vitro antiproliferative studies against human cancer cell lines to identify lead candidates with improved receptor affinity and anticancer potential. The invention further provides pharmaceutical compositions comprising the disclosed compounds for inhibiting VEGFR-2-mediated angiogenesis and abnormal cellular proliferation, thereby providing promising lead molecules for targeted anticancer drug discovery and therapeutic development.
