Abstract: The present invention relates to a novel adamantane-thiophene based Schiff base ligand, namely (E)-N- (adamantan“l-yl)-5-(l-(2-iiicoLinoylhydrazono)- ethyl)thiophene-2-carboxamide, and transition metal complexes thereof, and processes for preparation. The ligand is synthesized by condensation of 5-acetyl- N- (adamantan-l-yl)thiophene-2-carboxamide with nicotinic hydrazide in ethanol in the presence of acetic acid. The invention further provides Cu(II), Ni(II), and Co(TI) complexes obtained by coordination of the ligand with corresponding metal salts through azomethine nitrogen and carbonyl oxygen donor atoms. The compounds are characterized using elemental analysis, FTIR, UV-visible spectroscopy, NMR, mass spectrometry, ESR, cyclic voltammetry, thermogravimetric analysis, X-ray diffraction and density functional theoiy studies. The synthesized ligand and metal complexes exhibit significant antibacterial activity against Gram-positive and Gram- negative bacteria and demonstrate anticancer activity against cultured cancer cells. The invention provides potential pharmaceutical and therapeutic compositions comprising the ligand and its metal complexes for antimicrobial and anticancer applications.
