Abstract: The present invention relates to a nanoemulsion-loaded transdermal patch comprising Posaconazole for controlled and sustained antifungal drug delivery. Posaconazole is incorporated into a stable nanoemulsion containing oil phase, surfactant, co-surfactant, and aqueous phase to improve solubility and skin permeation. The nanoemulsion is embedded into a matrix-type polymeric patch comprising HPMC K100M and Eudragit RS100 using solvent casting. Multiple nanoemulsion and patch batches are optimized to obtain the best formulation. The optimized formulation comprises nanoemulsion batch NE-3 and patch batch P-3, providing sustained release for 24–72 hours, enhanced permeation, improved bioavailability, reduced gastrointestinal side effects, and better patient compliance. The invention provides a novel, stable, and commercially viable antifungal transdermal therapeutic system.
