Abstract: ABSTRACT A MULTIMODAL MICROSCOPY SYSTEM AND METHOD FOR WIDEFIELD FLUORESCENCE, SINGLE MOLECULE LOCALIZATION, AND BEAM-SCANNING IMAGING A multimodal microscopy system (100) and method (200) comprising a first and second laser source (1, 2) to generate excitation beams. A laser combining assembly (3) combines said first and second excitation beams, and the beam is collimated on a first dichroic beam splitter (DBS1) configured to separate said beam into first and second excitation beams. The second excitation beam is directed to a second dichroic beam splitter (DBS2) via said beam scanning unit (5). A combine beam of said first and second excitation beams from said second dichroic beam splitter (DBS2) falls on said beam splitting unit (9) which directs said combine beam toward said sample plane (10) and a camera (12) acquires images corresponding to the plurality of scanned spatial positions using the said second excitation beam during beam-scanning and also captures a plurality of images corresponding to widefield and single molecule localization.
