MEENAKSHI ACADEMY OF HIGHER EDUCATION AND RESEARCH 2. MEENAKSHI AMMAL DENTAL COLLEGE AND HOSPITAL ABSTRACT The present invention discloses a multi-responsive biopolymeric nano-system for simultaneous infection diagnosis and targeted therapeutic delivery. The nano-system comprises a biodegradable and biocompatible tamarind kernel polysaccharide (TKP) matrix crosslinked using calcium ions or natural cross-linkers to form a stable polymeric network. Functional linkages comprising pH-sensitive bonds, enzyme-cleavable bonds, and microbial metabolite-responsive moieties impart infection-responsive characteristics to the nano-system. Therapeutic agents encapsulated within the TKP matrix are released in a controlled manner upon exposure to infection-associated stimuli. Analytical agents incorporated within the matrix generate detectable signals indicative of infection through colorimetric, fluorescent, optical, or electrochemical responses. Pathogen-targeting ligands functionalized on the nano-system facilitate preferential localization at infection sites. The coordinated interaction of targeting, diagnostic, and therapeutic components enables simultaneous infection detection and localized treatment. The nano-system may be formulated as nanoscale particles, hydrogel dressings, microneedle patches, sprayable formulations, or implantable constructs.
