Abstract: The present invention provides a multi-layered mucoadhesive buccal patch for the modified-release, unidirectional transmucosal delivery of Berberine, Mangiferin, and Epigallocatechin gallate (EGCG). Prepared via sequential solvent-casting, the delivery system comprises 1–40% (w/w) active ingredients integrated into a mucoadhesive matrix (HPMC K4M, Carbopol 934P, PVP K30, and a plasticizer), which is bonded to an impermeable backing layer (Ethyl cellulose and dibutyl phthalate) constituting the remaining 60–99% (w/w). The structurally intact patch circumvents hepatic first-pass metabolism, maintains a physiological surface pH (5.5– 7.5), and yields a controlled-release profile characterized by an initial dissolution lag phase followed by sustained transmucosal release peaking between 60 and 90 minutes.
