Abstract: [047] The present invention relates to an aqueous mucoadhesive film-forming nasal spray composition intended to provide a temporary physical barrier on a nasal mucosal surface against fragrance-associated airborne irritants. The composition comprises hydroxypropyl methylcellulose (1.25% w/w), xanthan gum (0.10% w/w), glycerol (1.0% w/w), sorbitol (0.50% w/w), a pharmaceutically acceptable buffer in a quantity sufficient for pH adjustment and purified water in a quantity sufficient to complete the formulation. Hydroxypropyl methylcellulose provides film-forming and mucoadhesive characteristics; xanthan gum modifies rheology; glycerol contributes plasticisation and moisture retention; and sorbitol contributes moisture retention and tonicity adjustment. Following spray deposition, the formulation spreads over an accessible nasal mucosal region and develops a temporary hydrated polymeric coating as the aqueous vehicle redistributes or partially evaporates. The coating is intended to reduce direct contact of the coated mucosal surface with inhaled fragrance-associated irritants while permitting normal nasal airflow. The invention also relates to a method of preparing the composition by polymer hydration, mixing, excipient incorporation, pH adjustment and filling into a nasal spray container.
