Abstract: The present invention a microfluidic screening system (100) is disclosed for parallel preparation, characterisation, in-vitro evaluation, and technical ranking of exosome formulations. The system comprises a microfluidic isolation module (200), a cargo-loading module (300) having parallelised loading chips (400), a ligand conjugation chamber (500), a readout module sensor (600), a cell uptake assay plate (700), a processor-implemented ranking module (800), and a combinatorial matrix controller (900). The isolation module (200) enriches extracellular vesicles from donor-cell-conditioned medium, while the cargo-loading module (300) introduces cargo under controlled loading energy and temperature. The readout module sensor (600) generates particle-size, yield, fluorescence, and cargo-retention data, and the cell uptake assay plate (700) generates uptake and viability data. The ranking module (800) produces a lane-specific score and communicates through a hardware control interface to adjust a physical formulation parameter only when predetermined quality thresholds are satisfied, thereby enabling traceable, closed-loop, sterile, and reproducible formulation screening without therapeutic administration.
