Abstract: A METHOD OF ENZYME IMMOBILISATION USING KINETIC SAND AND CATIONIC PEPTIDE FOR FLOW BIOCATALYSIS. The present invention discloses a simple, eco-friendly, and reusable method for enzyme immobilization using kinetic sand as a porous support matrix. Kinetic sand comprises approximately 98% fine sand and 2% polydimethylsiloxane (PDMS), wherein the PDMS-coated sand particles enable enzyme immobilization through non-covalent interactions, including hydrophobic interactions and van der Waals forces, without requiring additional chemical linkers. The neutral pH and high porosity of the matrix allow efficient substrate diffusion and preservation of enzymatic activity. In one embodiment, free enzymes are immobilized directly onto kinetic sand and employed in a packed bed reactor for continuous flow catalysis. In another embodiment, kinetic sand is combined with moringa seed powder to enhance enzyme binding, stability, and reusability. The invention further provides methods for preparing enzyme immobilization matrices containing fine sand and silicone oil, with or without moringa seed powder. The immobilized enzymes demonstrate sustained catalytic performance over multiple reaction cycles, enabling two to three reuse cycles without loss of activity. The disclosed system offers advantages of low cost, mechanical stability, ease of handling, and environmental compatibility, making it suitable for applications in biocatalysis, wastewater treatment, and sustainable industrial processing.
