Abstract: The present invention relates to a validated liquid chromatography-tandem mass spectrometry (LC–MS/MS) method for the quantitative determination of oseltamivir in rabbit plasma. The method comprises isocratic chromatographic separation of oseltamivir on a reverse-phase C18 column using a mobile phase comprising 0.1% formic acid, acetonitrile, and methanol in the ratio of 10:70:20 (v/v/v) at a flow rate of 0.70 mL/min, followed by detection using positive electrospray ionization with multiple reaction monitoring, employing abacavir as an internal standard. Rabbit plasma samples are processed by acetonitrile-based protein precipitation, n-hexane partitioning, evaporation, and reconstitution prior to injection. The method is validated for selectivity, sensitivity, linearity, precision, accuracy, recovery, matrix effect, dilution integrity, carryover, and stability, and quantifies oseltamivir over the concentration range of 0.01 to 200 ng/mL with a lower limit of quantification of 0.01 ng/mL. The disclosed method provides a rapid, sensitive, selective, and reproducible bioanalytical tool suitable for preclinical pharmacokinetic, bioavailability, and bioequivalence investigations of oseltamivir.
