Abstract: The present invention relates to a topical antifungal composition comprising sterilized micronized barytes mine tailings and a lactic acid-containing biological penetration enhancer for treatment of fungal nail infections, particularly onychomycosis caused by Trichophyton rubrum. The invention provides an environmentally sustainable biomedical utilization of low-grade barytes mine waste generated from the Mangampet Barytes deposits in Andhra Pradesh. The mine tailings are pulverized, sieved to obtain particles smaller than approximately 0.01 mm, sterilized, and mixed with sour curd or another lactic acid-containing material, preferably in a weight ratio of about 10:1, to produce a topical paste capable of enhanced penetration into keratinized nail tissue. Experimental evaluation using infected toenail clippings demonstrated that the disclosed formulation significantly reduced fungal colony-forming units compared with untreated controls and aqueous barytes formulations, while exhibiting antifungal activity approaching that of a commercial amorolfine formulation. The invention simultaneously addresses environmental management of mining waste and development of an economical topical antifungal treatment with improved nail penetration characteristics. The composition may be formulated as a paste, gel, cream, ointment or other pharmaceutically acceptable topical dosage form for treatment or management of dermatophyte-induced nail infections.
