Abstract: The present disclosure proposes a method for preparing dual-metal functionalized three-dimensional chitosan-based scaffolds incorporating ionic crosslinking and coordinated metal-ion functionalization to provide porous biodegradable scaffold architectures suitable for tissue engineering and biomedical applications. The method comprises preparing a homogeneous chitosan solution in an aqueous acetic acid solution, casting the solution into a mould, freezing and lyophilizing the cast solution to obtain a porous three-dimensional chitosan scaffold (208), ionically crosslinking the scaffold using a sodium tripolyphosphate solution, preparing a dual-metal ion solution comprising zinc ions together with at least one secondary metal ion selected from cobalt ions and manganese ions, immersing the ionically crosslinked scaffold in the dual-metal ion solution to incorporate the metal ions within the scaffold matrix, and drying the scaffold to obtain the dual-metal functionalized three-dimensional chitosan-based scaffold exhibiting structural stability, controlled biodegradation, antibiofilm activity, cytocompatibility, and hemocompatibility.
