Abstract: The present disclosure proposes a method for detection, separation, and structural characterization of degradation impurities of Sebetralstat using chromatographic and mass spectrometric techniques. The method employs reverse-phase high-performance liquid chromatography (RP-HPLC) coupled with liquid chromatography–tandem mass spectrometry (LC-MS/MS) for identifying and characterizing degradation impurities formed during stress degradation of Sebetralstat. The method enables rapid separation and detection of Sebetralstat and its degradation impurities using a stability-indicating RP-HPLC system with optimized chromatographic conditions. The method facilitates accurate structural characterization of degradation impurities through the coupling of chromatographic separation with LC-MS/MS based mass spectral and fragmentation analysis. The method evaluates the stability of Sebetralstat under multiple forced degradation conditions, including acidic, alkaline, oxidative, thermal, photolytic, reductive, and hydrolytic environments.
