Abstract: The present invention discloses a method for green synthesis and anticancer evaluation of Costus comosus-derived silver nanoparticles for hepatocellular carcinoma chemoprevention. Aqueous leaf extract of Costus comosus is mixed with silver nitrate at eighty-five degrees Celsius to synthesize CC-AgNPs, confirmed by surface plasmon resonance at 463 nm. Characterization by ATR-FTIR, SEM, EDX, XRD, and HR-TEM reveals spherical morphology, elemental silver purity, and face-centred cubic crystalline structure with particle sizes of 5 to 12 nm. In-vitro evaluation in HepG2 cells demonstrates cytotoxicity, caspase-3 activation, Bax and p53 upregulation, and Bcl-2 downregulation confirming intrinsic apoptosis pathway activation. In-vivo evaluation in diethylnitrosamine-induced hepatocellular carcinoma demonstrates dose-dependent reduction in nodule incidence, restoration of hepatic enzymes, antioxidant improvement, tumour marker reduction, and histopathological recovery, providing improved anticancer efficacy, enhanced safety, and a sustainable green nanoplatform for hepatocellular carcinoma management.
