Abstract: A Media Composition and a Process for Fucoxanthin Pigment Production from a Tropical Marine Microalga The present invention discloses a culture medium and a process for improving the yield of a bioactive pigment fucoxanthin from the isolated strain of microalga Nitzschia sp. having accession number BDU009. The microalga was grown in controlled laboratory conditions of low light intensity (100 µmol m-2 s-1), at 15 ppt salinity, pH 7.8, 12h:12h photoperiod, in a modified f/2 culture medium. The culture medium contained the exogenous plant hormone, salicylic acid which was added at the beginning of exponential phase (3rd day) and concentrations of nitrate and silicate (6x and 4x the base concentration of f/2), which were added on inoculation day while preparing media and re-added on day 5 as required due to rapid consumption by actively growing and multiplying cells for metabolism and secondary metabolite production. A high concentration of fucoxanthin (8.3 mg g-1 dry weight) was obtained in the exponential culture phase (7 to 8 days) as assessed through High Performance Liquid Chromatography. The extracted pigment was chemically characterized via LC-MS and NMR to confirm the structure and purity of fucoxanthin. The invention can promote the growth of the Nitzschia sp. as well as enhance the fucoxanthin production in a short span. Overall the invention includes economical and less laborious upstream and downstream processing for obtaining enhanced fucoxanthin yield.
