Abstract: The present invention relates to a loop-mediated isothermal amplification (LAMP) diagnostic formulation for rapid detection of plant viruses, particularly Tomato Brown Rugose Fruit Virus (ToBRFV), and a process thereof. The formulation comprises an optimized isothermal amplification buffer, deoxynucleotide triphosphates (dNTPs), magnesium sulfate, betaine, a strand-displacing DNA polymerase, a colorimetric indicator, nuclease-free water, and a six-primer formulation comprising forward outer (F3), backward outer (B3), forward inner (FIP), backward inner (BIP), loop forward (LF), and loop backward (LB) primers configured to recognize conserved regions of the viral replicase gene. The process comprises standardized preparation of reagent solutions, controlled formulation under nuclease-free conditions, aliquoting, and storage to provide reproducible diagnostic performance. The formulation enables rapid isothermal amplification with direct visual detection without thermal cycling equipment, thereby providing a standardized, scalable, and field-deployable platform for reliable plant virus diagnosis.
