Abstract: ABSTRACT: The present invention relates to a ligand-functionalized nanostructured lipid carrier pharmaceutical composition for the treatment of atopic dermatitis. The composition comprises a first population of nanostructured lipid carriers containing Sulindac, a second population of nanostructured lipid carriers containing Mycophenolate Mofetil, and Indole-3-acetonitrile physically associated with the surfaces of the respective nanocarriers. Each nanostructured lipid carrier comprises a lipid matrix formed from at least one solid lipid, at least one liquid lipid, and a pharmaceutically acceptable surfactant. Sulindac is employed to modulate the aryl hydrocarbon receptor-associated pathway and support epidermal barrier function, whereas Mycophenolate Mofetil suppresses proliferation of T and B lymphocytes involved in inflammatory responses. The coated nanocarriers provide high drug entrapment, nanoscale particle size, acceptable colloidal stability, and sustained drug release for up to 72 hours. The invention further relates to a microemulsion-based preparation process comprising emulsification, high-speed homogenization, cooling, Indole-3-acetonitrile coating, and optional lyophilization.
