Abstract: ABSTRACT 5 A HYDROGEL PRECURSOR COMPOSITION FOR EXPANSION MICROSCOPY AND METHODS THEREOF The present disclosure provides a hydrogel precursor composition for expansion microscopy, comprising: (a) a photocrosslinkable polymer comprising poly(alkylene glycol) (multi)acrylate; (b) an ionic monomer comprising an acrylate 10 or methacrylate salt of a metallic ion; and (c) a photoinitiator; wherein the photocrosslinkable polymer and the ionic monomer are in a molar ratio range of 1:200 to 1:500. The present disclosure further provides a crosslinked hydrogel, a method of preparing biological sample and an expanded biological sample for expansion microscopy.
