Abstract: The present invention relates to a validated High-Performance Thin Layer Chromatographic (HPTLC) method for the quantitative estimation of Pramiracetam in bulk drug and pharmaceutical dosage forms. Chromatographic separation is achieved on silica gel 60 F254 precoated HPTLC plates using a mobile phase comprising methanol, toluene and triethylamine in a ratio of 8.0:1.8:0.2 (v/v/v). The chromatographic chamber is saturated for 25 minutes and densitometric scanning is performed at 263 nm. Under the optimized chromatographic conditions, Pramiracetam exhibits a compact and symmetrical chromatographic peak with an Rf value of approximately 0.52 ± 0.02. The method is linear over the concentration range of 200–3200 ng/band with excellent precision, accuracy, specificity, robustness and sensitivity. The method provides a rapid, economical and reproducible procedure suitable for routine quality control, assay determination, validation studies and pharmaceutical quality assurance in accordance with ICH guidelines.
