Abstract: The present invention relates to a novel microparticle-based herbal nasal spray formulation comprising extracts of Zingiber officinale and Ocimum sanctum for rapid relief of migraine. The formulation is designed for intranasal administration to facilitate fast systemic absorption and potential nose-to-brain delivery of bioactive phytoconstituents, thereby bypassing hepatic first-pass metabolism and enabling a quicker onset of therapeutic action compared to conventional oral dosage forms. The phytoconstituents present in Zingiber officinale, including gingerols and shogaols, exhibit anti-inflammatory, analgesic, and anti-emetic properties, while Ocimum sanctum provides neuroprotective, antioxidant, adaptogenic, and vasomodulatory effects through compounds such as eugenol, ursolic acid, and rosmarinic acid. The synergistic combination of these herbal extracts contributes to effective management of migraine symptoms by reducing neurogenic inflammation, modulating neurotransmitter activity, and alleviating associated nausea and stress. The formulation further comprises pharmaceutically acceptable excipients to optimize pH, viscosity, spray characteristics, stability, and mucosal compatibility. Evaluation studies including physicochemical characterization, in vitro drug release, ex vivo permeation, and nasal irritation assessment demonstrate safety, stability, and effectiveness of the developed formulation. The invention provides a safe, non-invasive, and patient-compliant herbal therapeutic alternative for rapid migraine management.
