Abstract: The present invention relates to a green process for extraction of omega-3 fatty acid-rich algal oil from Schizochytrium sp. using a natural deep eutectic solvent (NADES) system . The process comprises preparing a NADES comprising choline chloride and lactic acid, mixing the NADES with Schizochytrium sp. biomass to form a biomass suspension, and subjecting the biomass suspension to ultrasonication or microwave treatment. The treated biomass suspension is subsequently subjected to thermal treatment, followed by addition of hexane for lipid partitioning. A lipid-containing organic phase is separated by centrifugation and collected. The organic solvent is removed by rotary evaporation to obtain algal oil. The extracted oil comprises omega-3 fatty acids including docosahexaenoic acid (DHA), docosapentaenoic acid (DPA), and eicosapentaenoic acid (EPA). The process integrates NADES-assisted extraction, energy-assisted cell disruption, thermal treatment, phase separation, and solvent removal within a single extraction workflow .
