Abstract: The present invention relates to a feeder-free immunotherapeutic strategy for adoptive cellular therapy, particularly for the treatment of hepatocellular carcinoma. The invention provides a method for generating a heterogeneous lymphocyte composition comprising activated T lymphocytes and natural killer (NK) cells expanded ex vivo from a donor-derived mononuclear cell population. Immune effector cells are isolated using affinity-based techniques and cultured in a defined cytokine environment comprising interleukin-21 and interleukin-15, without reliance on feeder cells, interleukin-2, or anti-CD3 stimulation. The culture conditions enable concurrent expansion and activation of both innate and adaptive immune cell populations, resulting in a synergistic cytotoxic profile against tumor cells. The expanded heterogeneous lymphocyte composition demonstrates functional immune activation and is formulated for adoptive administration to cancer patients. The invention addresses limitations of existing single-cell-type immunotherapies by providing a scalable, reproducible, and biologically integrated cell therapy platform suitable for solid tumors, particularly hepatocellular carcinoma.
