Abstract: The invention disclosed an ocular drug delivery system in the form of a dual-loaded, biodegradable nanofiber film for the treatment of allergic conjunctivitis. The film features a unique nano-in-nano architecture comprising nanoparticles embedded within a matrix of nanofibers. The Invention disclosed the nanoparticles are formed from poly(lactic-co-glycolic acid) (PLGA) and encapsulate an antihistamine, such as azelastine hydrochloride. These drug-loaded nanoparticles are embedded within nanofibers formed from polyvinyl alcohol (PVA), which are co-loaded with an anti-inflammatory agent, such as ketorolac tromethamine. An ocular drug delivery allows for the simultaneous and sustained release of both therapeutic agents when the film is topically applied to an ocular surface. The invention provides a non- invasive, patient-friendly method for treating allergic conjunctivitis, overcoming the limitations of conventional eye drops by enhancing drug bioavailability and reducing dosing frequency.
