Abstract: A DUAL DRUG-LOADED NANOFIBER FILM FOR ENHANCED DRUG DELIVERY SYSTEM AND A PROCESS THEREOF The present invention relates to dual drug-loaded electrospun nanofibrous transdermal drug delivery system for controlled and sustained delivery of antidiabetic drugs. The system comprises Metformin Hydrochloride and Sitagliptin Phosphate incorporated into a hybrid polymeric matrix of PVA and Eudragit RS100. A homogeneous precursor solution containing the polymers, drugs, and solvent is processed by electrospinning to produce a highly porous nanofibrous scaffold with uniform fiber morphology and high drug entrapment efficiency. PVA promotes rapid hydration and initial drug diffusion, while Eudragit RS100 controls polymer swelling and drug permeation, enabling sustained drug release. The developed scaffold enhances transdermal permeation, improves bioavailability, reduces dosing frequency, and enhances patient compliance compared to conventional oral formulations. The invention also provides a reproducible manufacturing process with optimized electrospinning parameters for large- scale production of a stable transdermal patch. The platform offers an effective alternative for Type II Diabetes Mellitus management and sustained transdermal drug delivery.
