Abstract: The invention relates to portable optical diagnostic imaging for lateral flow assays, paper-based analytical devices, and microfluidic biosensors. Disclosed is a smartphone-assisted device having an external black enclosure, a matte black internal chamber, a 365 nm UV LED excitation module, an adjustable focusing optical tube with a biconvex focusing lens, a polarization optical filter, a strip or sample holder, a smartphone viewing window, and an electronic power system with rechargeable battery module, direct DC power input, driver circuit, heat dissipation unit, and protective housing. The arrangement maintains a fixed excitation and imaging path, suppresses stray and reflected light, and improves fluorescence signal capture and repeatability compared with conventional open or loosely aligned smartphone imaging setups. The method includes illuminating an assay region, filtering emitted light, capturing the image with a smartphone camera, and performing qualitative or quantitative analysis, principally for point-of-care fluorescence-labelled and colorimetric biosensor testing.
