Abstract: The present invention is a low-cost, kit-based method and formulation for extracting deoxyribonucleic acid (DNA) from bacterial cells and microbial biofilms, including both Gram-positive and Gram-negative bacteria. For cell resuspension, enzyme inhibition, cellular disruption, contaminant separation, and DNA precipitation, the process uses Tris-EDTA buffer, sodium dodecyl sulfate, Proteinase K, phenol-chloroform, sodium acetate, and isopropanol. For use in subsequent molecular biology applications, the technique effectively recovers genomic DNA while providing DNA having suitable yield and purity for subsequent molecular biology applications. Microbial cells are cultured, harvested and resuspended, treated with specific reagents, phase separated, precipitated, washed, and resuspended, and then the concentration and purity of DNA are measured using spectrophotometry. Biofilm, bacterial, oral, and dental swab samples, as well as associated genetic analyzes, can all be performed with this kit.
