Abstract: ABSTRACT A Composition and Method for Smart Multi-Stimuli Responsive Supramolecular Dual-Network Hydrogel Drug Delivery System The present invention relates to a smart supramolecular hydrogel composition and method for controlled drug delivery and biomedical applications. The hydrogel comprises a dual-network architecture integrating tragacanth gum, gelatin, and grafted poly(acrylamide) through combined covalent and reversible non-covalent interactions, forming a dynamic and self-adaptive matrix. The system exhibits multi-stimuli responsiveness, including pH, temperature, and enzymatic triggers, enabling site-specific and programmable drug release. Nano-carrier embedded domains facilitate staged or sequential delivery of therapeutic agents, while gradient porosity and anisotropic swelling enhance diffusion and tissue interaction. The hydrogel further demonstrates self-healing capability, biocompatibility, hemocompatibility, mucoadhesion, antioxidant activity, and biodegradability. In certain embodiments, artificial intelligence and machine learning models enable predictive and closed-loop modulation of drug release based on physiological conditions. The disclosed system provides improved drug loading, controlled release kinetics, and personalized therapeutic delivery, making it suitable for applications in drug delivery, tissue engineering, and regenerative medicine.
