Abstract: [067] A co-amorphous solid dispersion composition and method for preparation thereof for improving solubility and stability of active pharmaceutical ingredients (APIs) are disclosed. The composition comprises at least one API in an amorphous form, at least one co- former, at least one polymeric carrier, and one or more pharmaceutically acceptable excipients. The API and co-former are molecularly dispersed within an amorphous matrix to enhance aqueous solubility, dissolution rate, bioavailability, and storage stability compared to corresponding crystalline forms. The co-former and polymeric carrier stabilize the amorphous state and reduce recrystallization during storage. The method comprises selecting compatible components, preparing a precursor blend, processing the blend through solvent evaporation, spray drying, hot melt extrusion, freeze-drying, or combinations thereof, conditioning the obtained amorphous matrix, and formulating the matrix into a pharmaceutical dosage form. The disclosed composition is suitable for oral, injectable, and transmucosal drug delivery applications.
