Abstract: The present invention discloses a novel dual-drug nanoformulation for the therapeutic management of Type 2 Diabetes Mellitus (T2DM). The invention overcomes the technical challenge of simultaneously co-encapsulating a highly hydrophilic drug, Metformin (MET), and a highly lipophilic drug, Vitamin D3 (VD3), into a single delivery platform. The present invention utilizes a biodegradable chitosan-tripolyphosphate (CS-TPP) polymeric matrix synthesized via an optimized ionic gelation method. Crucially, the formulation integrates Pluronic F-127 (PF127) as a steric stabilizer to prevent phase separation of the contrasting therapeutics, ensuring high colloidal stability (+31.2 mV) and controlled drug release. The resulting VD3-MET-PF127-loaded CS-TPP nanoparticles exhibit an optimized core size of 150-265 nm. In vitro evaluations conducted on L6 myotubes confirm that this combined nanocarrier delivers synergistic biological outcomes, including enhanced glucose uptake, increased antioxidant enzyme activity, and improved mitochondrial health, all achieved at a significantly lower therapeutic concentration (62.5 µg/mL) compared to conventional single-agent drug formulations.
