Abstract: The present invention relates to a biodegradable nanoparticle-based pharmaceutical composition comprising chitosan-coated poly(lactic-co-glycolic acid) and polyethylene glycol-polycaprolactone hybrid nanoparticles co-encapsulating curcumin and paclitaxel for synergistic anticancer therapy. The hybrid nanoparticles exhibit mean diameters of 100-250 nanometers with encapsulation efficiencies exceeding 75 percent for both drugs. The chitosan coating provides pH-responsive release behavior with accelerated drug release at tumor acidic pH 5.0-5.5. The combination of curcumin-mediated NF-kB inhibition and P-glycoprotein efflux suppression with paclitaxel-induced microtubule stabilization achieves synergistic cytotoxicity with combination indices below 0.7 in MCF-7 and A549 cancer cell lines. In vivo studies demonstrate 83.1 percent tumor growth inhibition with reduced systemic toxicity.
