Abstract: A bioadhesive vaginal drug delivery formulation is disclosed for localized intravaginal administration of therapeutic agents. The formulation includes a therapeutic agent reservoir, mucoadhesive polymer network, hydration regulating component, pH modulating component, release modifying phase, and pharmaceutically acceptable carrier base. Upon contact with vaginal fluid, the formulation hydrates, adheres to the mucosal surface, and releases the active substance in a controlled manner. The system reduces leakage, improves residence time, supports vaginal pH compatibility, and is adaptable as a gel, film, insert, tablet, pessary, or applicator- dispensed dosage form.
