Abstract: ABSTRACT The present invention provides a three-dimensional bioprinted controlled drug delivery platform comprising pentoxifylline (PTX)-loaded liposomes and NIH 3T3 fibroblasts encapsulated within a GelMA hydrogel. The bioink comprises methacrylated gelatin, a biocompatible photoinitiator, PTX-loaded liposomes, and NIH 3T3 fibroblasts. The bioink is deposited as discrete droplets using an extrusion-based 3D bioprinter and photocrosslinked after deposition to form a structurally stable three-dimensional hydrogel construct. The liposomal phospholipid bilayer provides a first diffusion barrier for PTX, while the hydrated and porous GelMA network provides a second diffusion and degradation barrier, thereby enabling sustained drug release. The GelMA matrix further provides a cell-adhesive environment supporting fibroblast viability and proliferation. The resulting platform enables simultaneous evaluation of long-term cellular viability, scaffold stability, and controlled PTX release in a physiologically relevant three-dimensional environment
