Abstract: Abstract The present invention provides a novel method for preparing a detergent-based decellularized spinal cord extracellular matrix (ECM) scaffold from human, goat, porcine, and bovine spinal cord tissue. The method involves a two different decellularization method that includes perfusion and agitation with optimized detergent solutions concentration ranging from 0.5-5% to effectively remove cells while preserving the structural integrity and bioactivity of the spinal cord ECM. The resulting decellularized spinal cord ECM scaffold is biocompatible, supports cell adhesion, migration, and differentiation, and promotes functional recovery in the in vivo studies. It can be used for various tissue engineering applications, including spinal cord injury repair or central nervous system application.
