The application for the patent was published on September 11, 2026, under issue no. 37/2026.

Abstract: The present invention relates to a method for increasing the yield of exopolysaccharide (EPS) using the soil-isolated bacterial strain Weissella confusa EPSRMS.DYP1 through optimization of the production medium composition. Three different production media were formulated by varying the carbon source while maintaining a common basal composition containing peptone, yeast extract, disodium phosphate, and citric acid. Among the tested formulations, the medium supplemented with glucose and glycerol exhibited significantly enhanced EPS production compared to media containing individual carbon sources. The optimized medium produced EPS in the range of 0.7 to 0.85 g/100 mL of culture medium. The obtained EPS was extracted by alcohol precipitation and structurally characterized using Fourier Transform Infrared (FTIR) spectroscopy, confirming the presence of characteristic polysaccharide functional groups. Further, the EPS demonstrated prebiotic activity against beneficial probiotic strain Limosilactobacillus fermentum. Gas Chromatography-Mass Spectrometry (GC-MS) analysis confirmed the production of short-chain fatty acids during probiotic fermentation of the EPS, indicating its functional role in promoting gut microbial balance and intestinal health. The present invention therefore provides an efficient and scalable method for enhanced EPS production along with its application as a bioactive ingredient in nutraceutical, pharmaceutical, and functional food formulations.
