Abstract: ABSTRACT An integrated diagnostic imaging and computational framework is disclosed for the early detection and risk stratification of glaucoma via the quantitative analysis of retinal microvascular thinning. The system comprises an ultra-low-cost, portable handheld optical hardware assembly including a cylindrical camera body housing a 20-Diopter (20D) lens and an LED ring light, configured to mount onto a smartphone and capture high-resolution fundus images. Validated images are securely transmitted to a cloud server where a green-channel image is extracted and preprocessed via CLAHE. A specialized Gated U-Net deep learning model segments fine, high-curvature microvessels from background noise. The segmented vessels undergo morphological refinement and single-pixel skeletonization to accurately isolate the capillary network. The framework automatically calculates critical quantitative biomarkers, including vessel density, thickness, and connection health, to detect early-stage capillary dropout and generate structured clinical reports.
