Abstract: The invention relates to quinazolinone Schiff base derivatives comprising a 2- phenylquinazolin-4(3H)-one core substituted at the 3-position by an imine-linked aromatic or heteroaromatic group. Specific derivatives include compounds bearing pyridin-3-yl, phenyl, 4-nitrophenyl, 4-hydroxyphenyl and 2-chlorophenyl groups. A process is disclosed comprising benzoylation of anthranilic acid to form 2-benzamidobenzoic acid, cyclization to 2-phenylbenzoxazinone, reaction with hydrazine hydrate to obtain 3-amino-2-phenylquinazolin-4(3H)-one, and condensation with an aromatic aldehyde to obtain the corresponding Schiff base. The synthesized compounds were characterized using physicochemical and spectroscopic methods and evaluated using forced swim and tail suspension behavioural models. SGQ1, SGQ3 and SGQ5 showed reduced immobility times relative to control under the reported test conditions. In silico assessment indicated drug-like physicochemical profiles for the disclosed compounds. The derivatives are useful as research leads for further development of antidepressant agents.
