Abstract: The present invention discloses a rapid, high-throughput, and eco-friendly colorimetric method (designated as the DEEPTI Method) for the direct estimation, verification, and molecular differentiation of total amino functional groups and microbial biomass in biological matrices. The process utilizes a multi-purpose dual biochemical probe operating via a Proprietary Chromogenic Activator Reagent (PCAR) matrix combined with customized fluid extraction vehicles (PEC-X and PEC- Y) under an optimized 1-hour processing protocol. Spectrophotometric analysis across an extended short-wavelength range from 400 nm to 600 nm yields distinct spectral signatures, displaying an immediate colorimetric shift to deep Ruhemann’s Purple for Primary Amines and a highly reproducible Light-Yellow Complex for Secondary Amines (including L-Proline and Hydroxyproline). Validated under a strict Randomized Block Design (RBD), the method eliminates hazardous traditional procedures, demonstrates a statistically significant expansion in raw extraction yield, and yields a 99.43% reduction in operational testing expenses compared to commercial kits.
