Abstract: The present invention relates to the repurposing of colchicine, a marketed anti-gout and anti-inflammatory alkaloid, as an anti-urolithiatic agent. The invention integrates in silico target prediction, network pharmacology, molecular docking and molecular dynamics simulation to identify and validate carbonic anhydrase-2 (CA2; PDB 6B00) as a principal target of colchicine, with a binding affinity of -7.2 kcal/mol and confirmed structural stability over a 100 ns simulation. Colchicine is thereafter formulated as chitosan nanoparticles (C-CNP) by ionic gelation, yielding particles of mean size 115.3 nm with confirmed drug-polymer interaction by FTIR and an amorphous transition by XRD. Colchicine and C-CNP demonstrate dose-dependent inhibition of calcium oxalate crystal nucleation, aggregation and crystallization in vitro, and C-CNP exhibits a favourable biocompatibility and efficacy profile in MDCK renal cells (crystal-inhibition IC50 = 64.18 ± 0.04%; cell- viability IC50 = 35.58%). The invention thus establishes colchicine, particularly in nanoparticle form, as a mechanistically validated, cost-effective repurposed candidate for the prevention and treatment of urolithiasis.
