Abstract: The present invention relates to a personalized multi-omics digital kidney twin system powered by artificial intelligence for early-stage chronic kidney disease (CKD) management. The system comprises a data acquisition module configured to collect patient-specific genomic, transcriptomic, proteomic, metabolomic, renal imaging, wearable physiological, laboratory, and electronic health record data; a data processing module for feature extraction and harmonization; and a multi-omics integration engine configured to generate a unified biological representation of kidney health. A digital kidney twin modeling engine constructs a patient-specific nephron-level computational model using Graph Neural Networks (GNNs) and Physics-Informed Neural Networks (PINNs) to simulate renal physiological processes and disease progression. A predictive simulation module forecasts CKD onset, renal function decline, and treatment outcomes, while a reinforcement learning module identifies optimized preventive and therapeutic interventions. A federated learning framework enables privacy-preserving collaborative model training across healthcare institutions. The continuously updated digital kidney twin provides personalized risk assessment, disease progression forecasting, and clinical decision support for improved preventive kidney care. Fig 1 to 4.
